For successful islet transplantation it is important to input sufficient quantity of islets, hence it is required to know the volume of the transplanted islets. Several methods for estimating the volume from 2D microscopy images are used (the Ricordi table, spherical and ellipsoid models). The aim of this work is to evaluate the accuracy of these methods. With the help of Optical Tomography microscope we made 401 projections of individual islets and created 3D models by image reconstruction. We calculated the individual islet volumes by the isotropic Fakir method and compared them with results from the evaluated 2D methods for 65 real islets and 35 synthetically generated islets. Ellipse fitting method for islet volume estimation performed by plugins of Fiji software was shown to be the most suitable among the evaluated 2D methods.cze
