dc.description.abstract | Studium buněčného prostředí může odhalit možnosti využití nanočástic v biomedicíně. Stav buněčné membrány může být v korelaci s vitalitou buňky. Membrána, jako vnější obálka buňky, má polo-tekutý charakter s proteiny a molekuly lipidů, které jsou v neustálém pohybu. Předkládaná práce je zaměřena na tvorbu a ověření metodiky měření mechanických vlastností (Youngův modul, adheze) živých buněk. Mikroskopie atomárních sil (AFM) byla vybrána pro sledování mechanických vlastností buňky, konkrétně bylo použito módu silové spektroskopie (FS). Tato práce představuje dosažené výsledky porovnání pomocí různých tipů hrotů a ověření na několika typech buněk. Rozdílné povrchy byly vytvářeny pomocí potažení skla tenkou vrstvou, případně navázání konkrétních látek na měřící hrot - funkcionalizace hrotu. Práce pojednává o možnostech navázání EMC proteinu na měřící hrot a měření adheze membrány a proteinu. Testování se živými buňkami vyžaduje specifický přístup ve všech částech výzkumu. Výsledkem je porovnání souboru zdravých živých CT26 a 3T3 buněk se souborem fixovaných CT26 a 3T3. Velká část diskuze se věnuje samotné volbě hrotu, která je brána jako klíčová část nastavení daného experimentu. V neposlední řadě jsou v diskuzi pojednávány veškeré potíže, které při experimentu nastaly. Na základě zjištění z této práce bychom měli být schopni zjistit, zda daná choroba ovlivňuje samotnou membránu a naopak. | cze |
dc.description.abstract | The study of cell-environment can reveal the possibilities of using nanoparticles in biomedicine. The condition of the cell membrane can tell about the condition of the cell. The membrane as the outer envelope of the cell has a semi-fluid character, and the proteins and lipid molecules are in constant motion. The presented work is focused on creating and verification of methodology for measuring mechanical properties (Young's modulus, adhesion) of living cells. Atomic force microscopy was selected for exploration mechanical properties of a cell, force spectroscopy mod was used. This work presents achieved results of comparison using different tips of cantilevers and verification on several types of cells. Different surfaces are creating by coating of glass, or binding of specific substances to the measuring tip - the functionalization of the tip. The thesis discusses with the possibilities of a EMC protein binding on the tip of cantilever and measurement of adhesion of membrane and the protein. Working with living cells claims specific approach in all parts of the research. The result is a comparison of a set of healthy living CT26 and 3T3 cells with a set of fixed CT26 and 3T3 cells. A large part of the discussion deals with the choice of the tip, which is taken as the key part of the setup of the experiment. Last but not least, all the difficulties encountered during the experiment are discussed. Based on the findings from this work, we should be able to tell if the disease affects the membrane itself and vice versa. | eng |
