Úloha HLA antigenů u autoimunitních onemocnění

Abstract

Bakalářská práce na téma "Role HLA antigenů u autoimunitních onemocnění" se zabývá především objasněním souvislostí mezi vznikem autoimunitních onemocnění a přítomností specifických HLA antigenů. První část se zaměřuje na obecný popis HLA systému, jeho nomenklaturu a rozdělení do tříd. Rovněž je zde popsána funkce a dědičnost tohoto sytému. V závěru první části jsou rozebrány základní principy a metody využívané k typizaci HLA systému. Druhá část práce popisuje autoimunitu a podstatu autoimunitních onemocnění spolu se základními mechanismy, které udržují imunologickou toleranci vůči vlastním tkáním. S obecnými příčinami autoimunitních chorob je zde vysvětlen i vliv HLA antigenů na jejich rozvoj. V posledních kapitolách jsou zmíněny vybraná autoimunitní onemocnění, u kterých byla prokázána asociace s HLA antigeny. Praktická část se zabývá procesem typizace HLA systému konkrétně stanovením predispozičních alel podmiňujících rozvoj celiakie na dvou rozlišovacích úrovních. Jsou zde popsány jednotlivé kroky laboratorního procesu, jako je izolace DNA, PCR - SSP a také elektroforetické vyhodnocení. V rámci praktické části byla zpracována i statická data týkající se genotypizace HLA systému za účelem stanovení přítomnosti predispozičních alel ve vybraných laboratořích sídlících v České republice.

This bachelor thesis with a topic "Role of HLA antigens in autoimmune diseases" explains the relationship between specific HLA antigens and the development of autoimmune diseases. The first part describes basic information of the HLA system, its nomenclature and its division into classes. In this part, there is also the function and the hereditability of the HLA system mentioned. In the conclusion of this part, there is an analysis of basic principles with an analysis of serological and molecular-genetics methods used for HLA typing. The second part of this thesis describes autoimmunity and the nature of autoimmune diseases together with basic principles that maintain immunological tolerance to their own tissue. With general causes of autoimmune diseases, there is also the influence of HLA antigens explained. Lastly, some of the autoimmune diseases, whose development depends on specific HLA antigens, are mentioned in this part. The practical part of this thesis is devoted to the HLA typing procedure, especially to typing HLA alleles associated with celiac disease at two resolution levels. There is also a description of steps leading to genotyping; including DNA isolation, PCR - SSP, and electrophoretic evaluation. The conclusion of this part contains statistic data about genotyping HLA system in association with autoimmune diseases in some laboratories based in the Czech Republic.
