dc.description.abstract | Glykosidasy jsou hydrolytické enzymy, které lze při vhodné úpravě reakčních podmínek využít v laboratoři při syntéze oligosacharidů. Mezi glykosidasy řadíme enzymy mikrobiálního, rostlinného i živočišného původu. V rámci této práce jsou zkoumány ?- N- acetylhexosaminidasy patřící do skupiny glykosidas. V lidském organismu se účastní patofyziologie osteoartritidy nebo chronické myeloproliferativní nemoci. Jejich absence vede k závažným vrozeným poruchám metabolismu, jako je Tay-Sachs a Sandhoffova choroba, a v posledních studiích se prokázala i souvislost mezi mutacemi ?-N-acetylhexosaminidas a patofyzologii Alzheimerovy choroby. Výzkum těchto enzymů by mohl napomoci budoucí léčbě právě těchto onemocnění [1; 2; 3]. Cílem teoretické části práce byla literární rešerše enzymů glykosidas a využití genetického inženýrství ke změně jejich substrátové specifity. Praktická část práce zahrnuje PCR, izolaci a manipulaci s DNA, transformaci a kultivaci E. coli a P. pastoris, chromatografickou purifikaci jedné mutantní enzymové varianty a její biochemickou charakterizaci. V rámci této práce byla v hostiteli Pichia pastoris heterologně exprimována, produkována a charakterizována mutantní ?-N-acetylhexosamidasa z Talaromyces flavus, TfHex R218H. Druhá část práce je věnována analýze sekvencí genů mutantních ?-N- acetylhexosaminidas připravených řízenou evolucí s cílem identifikovat vnesené mutace. | cze |
dc.description.abstract | Glycosidases are hydrolytic enzymes that may be used for the laboratory synthesis of oligosaccharides by appropriate adjustment of reaction conditions. They are enzymes of microbial, plant and animal origin. In the frame of this work, we focus on ?- N- acetylhexosaminidases, belonging to the group of glycosidases. In the human organism, ?- N-acetylhexosaminidases may be involved in the pathophysiology of osteoarthritis or chronic myeloproliferative disease. The absence of ?-N-acetylhexosaminidases leads to severe congenital metabolic disorders, such as Tay-Sachs and Sandhoff's diseases, and recent studies have also shown a link between ?-N-acetylhexosaminidase mutations and the pathophysiology of Alzheimer's disease. Research on these enzymes might help to treat these diseases in the future [1; 2; 3]. The aim of the theoretical part of this work is a literature review of glycosidase enzymes and the use of genetic engineering to change their substrate specificity. The practical part includes PCR, isolation and DNA manipulation, transformation and cultivation of E. coli and P. pastoris, chromatographic purification of one mutant enzyme variant and its biochemical characterization. | eng |
