dc.description.abstract1.Expose fixed rat pancreatic tissue and/or cultred cells to several elements that are absent or only in trace amounts present in mammals (may include As, Ba, Cd, Cr, Pd, Hg, Se, Ag, Co, Cu) at appropriate concentrations and time to reach inter/intracellular concentrations ?100 ppm 2.Further fixate the exposed tissue in preparation for scanning electron microscopy (SEM) 3. Perform energy dispersive x-ray spectroscopy (EDx) of tissue 4.Analyze spatial localization of exposed element and evaluation of success and potential 5. Study to be performed at Cell Biology Department, University Medical Center Groningen, The Netherlands, under supervision of Dr. Ben Giepmanscze
dc.description.abstractFeature identification of high-resolution electron microscopy images can be improved with the addition of elemental detection via energy dispersive x-ray spectroscopy (EDX). Fixed rat pancreas samples were incubated in eleven different metallic compounds then elemental composition was analyzed with Phenom-World Pro-X tabletop scanning electron microscope with elemental analysis. From analysis of elemental localization, it was determined that pre-embedding incubation of tissue with heavy metals is a viable method for staining. The most promising metals: hafnium, gadolinium, and samarium, should be further explored for elemental labeling by EDX.eng
