Diamantu podobný uhlík(DLC) je metastabilní forma amorfního uhlíku obsahujícího spojení atomů uhlíku za pomoci sp2asp3hybridizovaných orbitalů. DLC vrstvy jsou polovodičesvysokou mechanickou tvrdostí, chemickou netečností, nízkým koeficientem tření aoptickou průhledností. Vtéto práci jsou popsány vlastnosti DLC vrstev, možnosti jejich uplatněnívortopedii, kardiologiiavdalších lékařských oblastech, jejich výhodyanevýhody, třídění podle poměru sp3/sp2 vazebametody stanovení tohoto poměru. Použitím mezivrstev či dopováním mohou být zlepšovány vlastnosti DLC. Dále byla studována biokompatibilita DLC vrstev připravených metodou pulsní laserové depozice. Vrstvysodlišnými fyzikálními vlastnostmi byly deponoványzterče tuhy, za použití různé hustoty energie laseru (od 4 do 11 J?cm -2). Byly připraveny vzorkysvysokýmanízkým poměremsp3/sp2vazeb. Pro vrstvy bylo naměřeno zastoupení grafitickýchadiamantových vazeb za pomoci spektroskopie Augerových elektronůafotoelektronů buzených rentgenovým zářením. Uvytvořených vzorků byl zjištěn vliv poměru sp3/sp2vazeb na jejich tvrdost, hrubost, kontaktní úhel, transmisi, zeta potenciálamorfologii povrchu. In vitro testy DLC vrstev byly provedeny použitím lidských keratinocytů afibroblastů. Sohledem na povrchové vlastnosti studovaných materiálů byla ohodnocena cytotoxicita, buněčná adheze, proliferaceaexprese profilových proteinů, jako jsou cytokeratinyafibronektin. DLC vrstvy byly studovány pro jejich následné uplatněnívmedicíně. Diamond-like carbon (DLC) isametastable form of amorphous carbon containing bonded carbon atoms of sp2and sp3hybridized orbital. DLC layers are semiconductors with highmechanical hardness, chemical inertness, low coefficient of frictionand optical transparency. In this work the characteristics of DLC layers, possibilities of applications in orthopedics, cardiovascular surgery together with other medical field, their advantages and disadvantages, the classification of DLC layers by ratio sp3/sp2bonds and methods for determination of this ratio are described. Properties of DLC layers can be improved by using interlayers or doping. Alsobiocompatibility of DLC thin films prepared by pulsed laser deposition method was studied. Layers with different physical properties were deposited fromthe graphite target, using various laser energy density (4 to 11 J/cm2). Layers of low and high sp3/sp2bonds ratio were prepared. The bonds were calculated using X-ray photoelectron and Auger spectroscopy. The generated samples were found to be influencedby the ratio sp3/sp2bonds ontheir properties such as roughness, hardness, Young's model of elasticity, contact angle, transmission, zeta potential and surface morphology of the atomic force microscope were measured. In vitro test of DLC layers was arranged using normal human keratinocytes and fibroblasts. Cytotoxicity, cell adhesion, proliferation and expression of profile proteins such a s keratins and fibronectin were evaluated with respect to surface properties of studied materials. DLC layers have been studied for their subsequent application in medicine.
