Tato práce se zabývá principem Mikroskopu atomárních sil (AFM) a popsáním dat naměřených na něm. AFM je způsobem měření, který dokáže stanovit topografii s vysokou přesností - v jednotkách délek atomů. Popis detekčního principu je hlavním tématem první části práce, kde jsou vyjádřeny vzájemně působící síly hrotu se vzorkem, a také jednotlivé měřící módy. Druhá část si klade za úkol vystihnout oblíbené trendy v biomedicíně, jako je čtení genetického kódu, zjištění vazební části u amyloidu nebo záznam TiO2 vrstev. Následuje stať, ve které je přiblížen samotný přístroj SPM Solver Next NT-MDT. Poslední a hlavní část práce je věnována problematice biokompatibilních tenkých vrstev, konkrétně TiO2 a hydroxyapatitu. Měření bude mít za cíl vystihnout závislost morfologie povrchu vrstev na depozičních podmínkách. Konkrétně se měřily nanodrsnosti Sq, Sa a výškový rozdíl mezi minimem a maximem Sy. Poslední část práce se zabývá okrajově vrstvou z hydroxyapatitu. The aim of this thesis is to describe the principle of the Atomic Force Microscope (AFM) works and how to deal with data, which can be measured on it. AFM is one of the measurement technologies, which could detect topography in very precise quantity units, as are atomic lengths. The first part ofthis thesis concerns with detecting principle, there are described acting forces between scanned sample and tip of AFM, the second point of the first part is description of various measuring modes. The second chapter of this thesis deals with todays most popular trends in biomedicine, such as reading of genetic code, finding binding structure of amyloids or record of dynamical process on cit membrane. The next and also main part of the thesis is focused on detection topography of thin films, specifically the samples of TiO2 and hydroxyapatit thin films. The practical part examines the dependence of the topography of film surface on deponation conditions. Nano-roughnesses Sq, Sa and height altitude between minimum and maximum Sy were measured, to be specific. The last part of my thesis concerns marginally with film of hydroxyapatite.
