The topic of this diploma thesis is application recombinant congenic strains of mice of CcS/Dem series for mapping of complex traits. I focused on use of these strains for study of genetic susceptibility to intracellular parasite Leishmania major. Determination of genes modifying susceptibility or resistance enables better understanding of mechanisms of not only this disease, but also another complex polygenic disease, for example cancer. Using these recombinant congenic strains, developed from susceptible strain BALB/cHeA and resistant strain STS/A, is possible to examine the modifying genes alone, because all of twenty CcS/Dem strains carries random part (12.5%) of genes of STS/A strain on the background of BALB/cHeA strain (87.5%). Comparing the results of genotypization with phenotype (patological and imunnological reactions) it is possible to determine candidate loci of susceptibility modifying genes. Main output of this diploma thesis was mapping of areas of STS origin in CcS/Dem strains, which leads to better understanding their structure and to improve the CcS/Dem strains as a tool for analyzing QTLs. I also took part in preparation of CcS/Dem recombinants, which are used for linkage analysis....

One of the most dynamically developing medical field is regenerative medicine. Its main goal is to promote tissue repair. The aim of this thesis was to implement and optimize a new automated capillary immunoassay system, the size-based Simple Western system. A typical marker ofchondrogenic differentiation'type II collagen production -was detected in nanofiber samples with seeded mesenchymal stem cells. Functionalized PCL/PVA nanofiber scaffolds with incorporated thrombocytes (natural source of growth factors) were prepared by emulsion electrospinning technique. The tested system preserved biological activity of the delivered growth factors and stimulated adhesion, proliferation and viability of the seeded MSCs throughout the experiment....

Methods ofanalysis ofgene expression, whichincludeRNA isolation, RT PCR (creating cDNA and real time PCR), and methods involving detection of loci influencing immunological parameters belong to a large family ofmethods in biomedical research. Mastering these techniques is essential for successful fulfillment of aims of my thesis. In the first part of my work I contributedto the determination of the three loci, whichhaveaffectatthe immunological parameters inuninfected mice. I performed DNA isolation and DNA typingin two independentexperiments, which together comprised 209 individuals belonging to the mouse F2hybrids between strains O20 and OCB-43. For typing were selected7 markers: D2Mit14, D2Mit37, D2Mit126, D3Mit51, D5Mit43, D15Mit5 and D15Mit13. Statisticalevaluation was performedbyANOVA in Statisticasoftware. We detected three loci on chromosomes 2, 3 and 5 influencing size of secondary lymphoid organsin uninfected mice. Locus on chromosome 5 has a major effect on the regulation of the number of cells in inguinal lymph nodes. Interactions between loci on chromosome 2 and 3 influences the relative weight of the spleen. In the second partof my thesis I studiedexpression offour genes: Synaptotagmin 14, Fcgamma receptor 4, Guanylate binding protein 1, Guanylate binding protein 5in lymph nodes of mice infected with Leishmania major. RNA samples were isolated from at least 6 infected and uninfectedmice from 12 different strains. The results of Real-time PCR were normalaiyed using Glyceraldehyde-3-phosphate dehydrogenaseas a house keeping gene. Differences in expression genes in infected and uninfected mice of the same strain or between diffrent strains were analzyed using STATISTICA program....
