Teoretická část bakalářské práce byla věnována literární rešerši týkající se tématu monoklonálních protilátek, a to zejména přípravě jejich konjugátů pro využití v diagnostice a terapii. V rámci rešerše o přípravě radioimunokonjugátů monoklonálních protilátek byly popsány metody konjugace s ligandy, vybrané typy chelatačních činidel a radionuklidy využívané v oboru teranostiky. Experimentální část bakalářské práce byla věnována optimalizaci přípravy konjugátů bevacizumabu a rituximabu a jejich následnému značení vybranými radionuklidy 68Ga a 161Tb. Optimalizace podmínek přípravy zahrnovala nalezení vhodné metody separace protilátek z jejich infuzních roztoků a stanovení optimálních podmínek pro jejich konjugaci s vybranými chelatačními činidly. Těmi byly zvoleny DOTA-NHS ester a p-SCN-Bn-DOTA. Při konjugaci bevacizumabu byly zkoumány tyto poměry látkového množství protilátka:ligand 1:10, 1:20, 1:50 a 1:100. Při konjugaci rituximabu s DOTA-NHS esterem to byly poměry 1:20, 1:50 a 1:100 a při konjugaci s p-SCN-Bn-DOTA poměry 1:10, 1:20 a 1:50. Připravené konjugáty byly poté značeny 68Ga a 161Tb. Při značení 68Ga byly porovnány všechny zmíněné poměry látkových množství a pro značení 161Tb byly následně použity poměry, které při značení 68Ga poskytly nejvyšší výtěžky. Pro konjugáty DOTA-NHS-bevacizumab a DOTA-NHS-rituximab to byl poměr 1:20 a pro konjugát DOTA-SCN-rituximab poměr 1:10. V případě značení bevacizumabu 68Ga a 161Tb se výtěžky značení pohybovaly v rozmezí 20-40 %. V případě rituximabu dosáhly pouze 5-20 %. Při přečistění radioimunokonjugátu od nezreagovaného radionuklidu byly porovnány metody separace pomocí HPLC a odsolovací kolony. Radioimunokonjugát [161Tb]DOTA-NHS-bevacizumab byl následně testován in vivo na myším modelu na linii SKOV3 a jeho distribuce v organismu byla pozorována pomocí SPECT zobrazování. The theoretical part of the bachelor's thesis was devoted to literature research related to the topic of monoclonal antibodies, in particular to the preparation of their conjugates for use in diagnosis and therapy. As a part of the research on the monoclonal antibodiess conjugates preparation, methods of conjugation with ligands, selected types of chelating agents and radionuclides used in the field of theranostics were described. The experimental part of the bachelor thesis was devoted to the optimization of the preparation of bevacizumab and rituximab conjugates and their subsequent labelling with selected radionuclides 68Ga and 161Tb. Optimizing the preparation conditions included finding a suitable method for separating antibodies from their infusion solutions and determining optimal conditions for their conjugation with selected chelating agents. The selected chelating agents were DOTA-NHS ester and p-SCN-Bn-DOTA. The following antibody:ligand mass ratios of 1:10, 1:20, 1:50 and 1:100 were investigated for bevacizumab conjugation. When rituximab was conjugated with DOTA-NHS ester, the ratios were 1:20, 1:50, and 1:100, and when with p-SCN-Bn-DOTA, the ratios were 1:10, 1:20, and 1:50. The prepared conjugates were then labelled with 68Ga and 161Tb. When labelling with 68Ga, all the mentioned ratios of substance amounts were compared, and for labelling with 161Tb, the ratios providing the highest yields when labelled with 68Ga were used. It was a 1:20 ratio for the DOTA-NHS-bevacizumab and DOTA-NHS-rituximab conjugates, and a 1:10 ratio for the DOTA-SCN-rituximab conjugate. In the case of bevacizumab 68Ga and 161Tb labelling, the yields ranged from 20-40%. In the case of rituximab, they reached only 5-20%. In the purification of the radioimmunoconjugate from the unreacted radionuclide, separation methods using HPLC and desalting columns were compared. The [161Tb]DOTA-NHS-bevacizumab radioimmunoconjugate was subsequently tested in vivo in a SKOV3 mouse model and its distribution in the organism was observed using SPECT imaging.
