Tématem bakalářské práce je toxoplazmóza u těhotných žen a metody její laboratorní diagnostiky. Bakalářská práce shrnuje dosavadní získané poznatky o prvoku Toxoplasma gondii (dále jen T. gondii) z kmene Apicomplexa, který způsobuje onemocnění zvané toxoplazmóza, patřící mezi nejčastější zoonózy v ČR i ve světě. V teoretické části práce je uvedena historie objevu T. gondii, životní cyklus zahrnující pohlavní i nepohlavní fázi prvoka a mechanismus působení v těle hostitele. Jsou zde popsány zdroje nákazy, kterými se může člověk infikovat a prevence před nimi. Dále je zde uveden průběh onemocnění a přímá i nepřímá laboratorní diagnostika T. gondii. V neposlední řadě jsou probrány možnosti léčby nemoci. V praktické části je podrobně popsán postup stanovení screeningové metody KFR pro stanovení celkových protilátek proti T. gondii a postup doplňujících metod EIA pro určení přesné třídy imunoglobulinů společně s aviditou IgG. Dále je zde statistické zhodnocení počtu vyšetření a výsledků metody KFR provedené ve FN Plzeň v letech 2011 až 2021. Ze zpracovaných dat bylo zjištěno, že počet stanovení na celkové protilátky proti T. gondii metodou KFR se dlouhodobě snižuje, i když počet pozitivních výsledků není výrazně snížen. Počet výsledků s vysokým titrem u těhotných žen se v roce 2021 významně snížil. Je tedy možné, že k onemocnění dochází dříve než ve fertilním věku, a tím nedochází k ohrožení plodu. The topic of the bachelor thesis is toxoplasmosis in pregnant women and methods of its laboratory diagnostics. The bachelor thesis summarizes the existing knowledge about the protozoan Toxoplasma gondii (hereinafter referred to as T. gondii) from the Apicomplexa strain, which causes a disease called toxoplasmosis, one of the most common zoonoses in the Czech Republic and the world. The theoretical part of the thesis presents the history of the discovery of T. gondii, the life cycle involving the sexual and asexual phases of the protozoan and the mechanism of action in the host's body. It describes the sources of infection through which a person can become infected and prevention from them. In addition, the course of the disease and direct and indirect laboratory diagnosis of T. gondii are presented here. Finally, the possibilities of treating the disease are discussed. The practical part describes in detail the procedure for the determination of the KFR screening method for the determination of total antibodies to T. gondii and the procedure of additional EIA methods for determining the exact class of immunoglobulins together with the IgG avidity. There is also a statistical evaluation of the number of examinations and results of the KFR method performed at the FN Plzeň between 2011 and 2021. From the processed data, it was found that the number of determinations for total antibodies against T. gondii by the KFR method decreases over the long term, although the number of positive results is not significantly reduced. The number of results with a high titre in pregnant women decreased significantly in 2021. It is therefore possible that the disease occurs earlier than in the childbearing age, and thus there is no threat to the fetus.
