dc.description.abstract | Ve své diplomové práci jsem se zaměřila na sledování změn mechanických a strukturních vlastností kolagenových nanovrstev vlivem činnosti buněk. Tyto změny byly sledovány během a po expozici v samotném kultivačním médiu a v přítomnosti dermálních fibroblastů a osteoblastů, které byly na vzorcích kultivovány po dobu tří týdnů. Úspěšnost kultivace byla hodnocena pomocí fluorescenční mikroskopie a pomocí hodnocení metabolické aktivity kultivovaných buněk. Změny mechanických vlastností byly měřeny jednoosou tahovou zkouškou, u které se vyhodnocoval počáteční modul pružnosti v prvním a posledním zatěžovacím cyklu a mez pevnosti kolagenové vrstvy. Strukturní změny byly sledovány pomocí infračervené spektroskopie a elektronové mikroskopie. Kultivace byla hodnocena jako úspěšná. Změny počátečního modulu pružnosti byly pozorovány nejvíce u prvního zatěžovacího cyklu, kde byly u vzorků s fibroblasty i osteoblasty zaznamenány statisticky významné rozdíly hodnot mezi 1. a 21. prvním dnem kultivace. U posledního zatěžovacího cyklu byly statisticky významné změny mezi 1. a 21. dnem kultivace pozorovány pouze u vzorků s osteoblasty. Naopak, mez pevnosti rostla jak v médium bez buněk, tak v přítomnosti obou typů buněk, a tuto změnu tedy nelze přičíst pouze působení buněk. Infračervená spektroskopie ukázala změny v helikální struktuře vzorků kultivovaných s osteoblasty. Na snímcích z elektronové mikroskopie, ze kterých byly měřeny průměry vláken, změny pozorovány nebyly. Fibroblasty i osteoblasty mohou svojí činností ovlivňovat mechanické a strukturní vlastnosti kolagenových nanovrstev. Popis buněčných mechanismů odpovědných za tyto změny bude součástí dalšího výzkumu, který bude zaměřen především na analýzu enzymových a jiných remodelačních aktivit fibroblatů a osteoblastů. | cze |
dc.description.abstract | In my thesis I focused on changes in mechanical and structural properties of collagen nanolayers due to cellular activity. These changes were monitored during and after exposure in the culture medium itself and in the presence of dermal fibroblasts and osteoblasts which were cultured on collagen nanolayers for three weeks. The cell culture on collagen nanolayers was assessed by fluorescence microscopy and metabolic activity. Changes in mechanical properties were measured by a unilateral tensile test in which the initial modulus of elasticity in the first and last load cycle and the ultimate tensile strength of the collagen layer was evaluated. Structural changes were monitored by infrared spectroscopy and electron microscopy. Fibroblasts and osteoblasts were succesfully plated and cultured on collagen nanolayers. Changes in the initial modulus of elasticity were observed most in the first load cycle, where both the fibroblast and the osteoblast samples showed statistically significant differences between the 1st and 21st day of the cultivation. For the last load cycle, statistically significant changes between 1st and 21st day of cultivation were observed only in samples with osteoblasts. On the other hand, ultimate tensile strenght increased in both cell culture media as well as in layers with cultured cells, thus the changes in ultimate tensile strength cannot be addressed only to cell activity. The infrared spectroscopy revealed the changes in helical structure of collagen nanolayers only in osteoblast specimens. Fiber diameters changes which were measured from images from electron microscopy were not observed. Fibroblasts and osteoblasts can influence mechanical and structural properties of collagen nanoparticles. Identification of cellular mechanisms responsible for the mechanical changes will be the aim of following research focused on the anaylsis of enzymatic activity and remodelation procedures of dermal fibroblasts and osteoblasts. | eng |
