dc.description.abstract | Normální kožní mikrobiom je přítomen na zdravé kožní tkáni. Za fyziologického stavu si zachovává svou typickou bakteriální skladbu, specifickou pro každého člověka. Předpokládá se, že změny v jeho složení mohou nastat v souvislosti s výskytem nádorové tkáně. Předkládaná práce si dává za cíl stanovit diverzity mikrobiomu a nalézt nové možnosti léčby melanomu. Tímto výzkumem se zabýval Ústav živočišné fyziologie a genetiky AV ČR, který ke sledování využil modelové organismy, konkrétně animální model liběchovských miniprasat. Celkem bylo odebráno 90 vzorků z povrchu melanomu, středu melanomu a kůže v jeho okolí u zvířat ve věku 6., 8., 10., a 12. týdnů. Pro účely této práce byly vyhodnoceny vzorky melanomové a kožní tkáně odebrané v 10. týdnu věku zvířete. K analyzování mikrobiomu byly využity metody molekulární biologie. Nejprve byla ze seškrabů obou typů tkání izolována DNA, která byla následně amplifikována pomocí PCR. Primery použité k amplifikaci ohraničovaly V3 oblast genu 16S ribozomální RNA. Úseky DNA byly rozděleny metodou denaturační gradientové gelové elektroforézy a po vizualizaci fragmentů bylo 48 z nich vybráno a komerčně osekvenováno. Nukleotidové sekvence vzorků byly zpracovány v programu Geneious a následně porovnány algoritmem BLAST s databází nukleotidových sekvencí genu pro 16S ribozomální RNA. Porovnáním získaných výsledků jsme zjistili diverzity bakteriálních druhů mezi vzorky kůže a melanomu. Melanomová tkáň vykazovala výrazně zvýšený výskyt bakterií Fusobacterium a Trueperella, které se fyziologicky na kůži nevyskytují. Průkazem těchto rodů na melanomu tak můžeme předpokládat, že se jedná o bakterie, jejichž výskyt je vázán na nádorovou tkáň. Zhodnocení mikroorganismů vyskytujících se na melanomové tkáni a kůži v jejím okolí by mohl znamenat posun při diagnostice nebo terapii tohoto druhu rakoviny kůže. | cze |
dc.description.abstract | Normal skin mikrobiom is present on healthy skin tissue. In physiological condition, retains its typical bacterial composition, identical for each person. It is assumed that changes in its composition can occur in relation to the occurrence of tumour tissue. The thesis aims to determine the diversity of microbiome and to find new possibilities of treatment of melanoma. This research was dealt with by the Institute of Animal Physiology and genetics of the ASCR, which used to monitor the model organisms, namely the model of Libechov Minipigs. In total, 90 samples were taken from the surface of melanoma, the centre of melanoma and the skin in the vicinity of animals aged 6, 8, 10, and 12 weeks. For the purpose of this work, the samples of melanoma and skin tissue collected in 10th week of age of the animal. Molecular biology methods were used to analyze microbiome. At first, DNA was isolated from the cutaneous scrapings of both types of tissue, which was subsequently amplification of PCR. Primers used to amplification specifically targeting V3 region of the 16S ribosomal RNA gene. DNA fragments were divided by the method denature gradient gel electrophoresis and after visualization of the fragments, 48 of them was selected and commercially sequenced. Nucleotide sequence samples were processed in the Geneious program and then compared by the BLAST algorithm with the nucleotide sequencing gene database for 16S ribosomal RNA. By comparing the results obtained we found the diversity of bacterial species between skin samples and melanoma. Melanoma tissue showed a significantly increased incidence of bacteria Fusobacterium and Trueperella, which are not physiologically present on the skin. The demonstration of these genera in melanoma can therefore assume that occurrence of these bacteria is bound to the tumor tissue. The evaluation of the microorganisms occurring on the melanoma tissue and the skin around it could mean a shift in the diagnosis or therapy of this type of skin cancer. | eng |
