Stanovení titru anti-A a anti-B u dárců krve krevní skupiny 0

Abstract

Předmětem bakalářské práce je zjistit procentuální podíl dárců krve krevní skupiny 0 s tzv. nízkým titrem protilátek anti-A a anti-B v plazmě. Titrace byla prováděna na analyzátoru Galileo NEO Iris a část z nich porovnána s výsledky manuálních metod - zkumavkovým testem a sloupcovou aglutinací. Teoretická část bakalářské práce se zabývá imunohematologií erytrocytů, kde jsou probrána především následující témata - erytrocytární antigeny, antierytrocytární protilátky a krevní skupiny s důrazem na AB0 a Rh systém. Kromě toho jsou zde popsány základní imunohematologické techniky. Dále jsou v práci popsány transfuzní přípravky, které jsou v současné době používány, předtransfuzní vyšetření prováděná před jejich aplikací a také základy účelné hemoterapie. V neposlední řadě jsou v práci také zmíněny potransfuzní reakce a komplikace, které mohou v důsledku podání transfuze nastat. Metodika práce popisuje základní principy a pracovní postupy u použitých metod - analyzátoru Galileo NEO Iris, zkumavkového testu a manuální sloupcové aglutinace. Ve výsledcích jsou naměřené hodnoty zpracované pomocí grafů a tabulek, nechybí ani jejich statistické zhodnocení, které poskytuje mj. informace o vzájemné korelaci použitých metod. Získané výsledky jsou porovnány s výsledky z jiných zemí světa.

The subject of the Bachelor's thesis is to find out the percentage of blood donors type O with low titer of anti-A and anti-B antibodies. The titration was made on the analyzer Galileo NEO Iris and some of them compared with the manual methods - the tube test and the column agglutination. Theoretical part of the Bachelor's thesis describes the immunohematology of the red blood cells, especially these topics: red blood cells antibodies and antigens and blood groups (mainly ABO and Rh systems). There are also described the basic immunohematological tests. Blood products, pre-transfusion testing and the principles of effective hemotherapy and post-transfusion reaction and complication are also described there. In the methods are the principles and the workflows of the used techniques - analyzer Galileo NEO Iris, tube test and column agglutination. The results of the Bachelor's thesis are compared using the diagrams and tables. There is also a statistic evaluation of the results, which also provides information on the correlation of the methods used. The obtained results are compared with the results from other countries.
