Jeden z cílů této práce je detailně popsat mechanismy poškození jaderné DNA z hlediska radiační fyziky a chemie, spolu s mechanismem vzniku mikrojader v lidských buňkách. Dále uvést přehled experimentálních studií, které se mikrojádrům věnují a příklady využití detekce mikrojader v biodozimetrii. Praktická část práce se zaměřuje na analýzu počítání mikrojader v lidských buňkách nasnímaných na fluorescenčním mikroskopu po označení fluorescenčním barvivem DAPI. Automatizace počítání mikrojader způsobených ozářením protonovým svazkem je důležitá pro urychlení výzkumu radiobiologie protonů, proto byl vytvořen program v programovacím jazyku PYTHON, který automaticky spojuje a vyhodnocuje snímky z mikroskopu. Cílem programu je spočítat nejen počet binukleárních buněk a mikrojader na sérii snímků, ale rovněž dokáže ke každé binukleární buňce přiřadit počet mikrojader které ji náleží. Tato studie shrnuje moderní metody používané pro analýzu poškození DNA. Snaží se pomoct v akceleraci výzkumu radiobiologie protonů pomocí programu pro automatickou analýzu vzorků a objasňuje některé problémy se zpracováním obrazu při tvorbě programu pro automatickou analýzu snímků. Navíc taky zhodnocuje účinnost protonového svazku pro indukci mikrojader v buněčných kulturách Daoy v závislosti na dávce. Na závěr bylo porovnáno počítaní binukleárních buněk a mikrojader pomocí programu a manuálně. Celková pravděpodobnost detekce binukleárních buněk byla stanovena na 89 procent a celková pravděpodobnost detekce mikrojader byla stanovena na 85 procent. One of the main objectives of this thesis is to thoroughly describe the mechanisms of the DNA damage from the physical and chemical point of view, together with the mechanism of micronuclei formation in human cells. Moreover, to append the summary of micronuclei oriented experimental studies and some examples of the micronuclei detection utilization in biodosimetry. The practical part of this thesis mainly focuses on micronuclei scoring of human cells scanned by a fluorescence microscope after marking them with the fluorescent DAPI stain. A program for automated analysis of micronuclei was created in the PYTHON programming language in order to automate the micronuclei scoring, which plays an important part in accelerating the research of the proton irradiation radiobiology. The main purpose of this program is not only to score the total number of binucleated cells and micronuclei in the data series also to assign the exact number of micronuclei to the given cell. This study summarizes the majority of methods for the DNA damage analysis. It is designed to help the acceleration of the proton radiobiology research through the program for the automated analysis of samples and it clarifies some of the issues which can emerge during the process of creating the program for automated sample analysis. Moreover, the dependence of the proton beam efficiency on the mironuclei induction in the Daoy cell line on the absorbed dose is included. In conclusion, the binucleated cells and micronuclei counting using the program developed were compared. The total binucleate cells detection probability was determined to 89 percent and the total micronuclei detection probability was determined to 85 percent.
