Cílem práce bylo navrhnout a implementovat nástroj pro automatické vyhodnocení obrázků DNA origami pořízené mikroskopií atomárních sil (AFM). Vyhodnocení mělo obsahovat úpravu obrázku vedoucí k jeho segmentaci a následné klasifikaci rozpadlých a úplných struktur DNA origami. Cílem klasifikace bylo také zhodnocení navázání nanočástic na strukturu DNA origami. Práce se zabývá syntézou vzorku DNA origami, jeho měření na AFM a jeho zpracování v prostředích ImageJ a Python. Výsledným produktem je jednoduchý detektor struktur DNA origami jak v celistvé, tak v rozpadlé podobě, a detektor nanočástic. Specificita klasifikačního algoritmu pro detekci rozpadlých struktur vyšla na 0,87 a senzitivita na 0,77. Senzitivita detekce nanočástic je 0,93. Vyvinutý algoritmus řeší problematiku ručního počítání struktur DNA origami na obrázku. The goal of the thesis was to design and implement a tool for automatic evaluation of DNA origami images taken by atomic force microscopy (AFM). The evaluation should include image processing leading to its segmentation and subsequent classification of broken and complete DNA origami structures. The aim of the classification was also to evaluate the binding of nanoparticles to the DNA origami structure. Thesis deals with the synthesis of a DNA origami sample, its measurement on AFM and its processing in the ImageJ and Python environments. The resulting product is a simple detector of DNA origami structures, both intact and broken, and a nanoparticle detector. The specificity of the classification algorithm for the detection of decayed structures was 0.87 and the sensitivity was 0.77. The sensitivity of nanoparticle detection is 0.93. The developed algorithm solves the problem of manual counting of DNA origami structures in the image.
