Cílem této bakalářské práce je podrobné seznámení se s principy klasické optické mikroskopie a s moderními technikami, které umožňují překročit klasickou Abbeho rozlišovací mez optických mikroskopů a dosažení tzv. superrozlišení. Úkolem je tedy provedení analýzy jednotlivých metod z hlediska jejich fyzikálního principu a konstrukce mikroskopů, srovnání rozlišení jednotlivých metod a jejich možných aplikací v praxi, zejména v oblasti biologie a medicíny. V následující kapitole je pro ilustraci představeno pár příkladů komerčně dostupných mikroskopů umožňující získat superrozlišení. Dále byla pomocí systému MATLAB provedena analýza vlivu aberací na rozptylovou funkci bodu. Nedílnou součástí této práce je přiložené CD obsahující elektronickou verzi bakalářské práce a počítačovou simulaci vlivu aberací na rozlišení optické soustavy. The aim of this bachelor thesis is to introduce the principles of the conventional microscopy in detail and to present modern techniques which enable us to break the Abbe resolution limit and to achieve the socalled super-resolution. The goal is to undertake the analysis of particular methods from the point of view of the physical principle and the construction of the microscopes, the comparison of the resolution of particular methods, their possible application in practice, especially in the area of biology and medicine. The next chapter introduces several examples as an illustration of commercially accessible microscopes which enable us to achieve the super-resolution. The analysis of the influence of the aberrations on the point spread function was done via the MATLAB system. The integral part of this work is the enclosed CD-ROM which includes the electronic version of the bachelor thesis and the computer simulation of the influence of the aberrations on the resolution of the optical system.
