Disertační práce se věnuje studiu dvou bílkovin-nové FAD-dependentní oxidore- duktáze z houby Chaetomium thermophilum (CtFDO) degradující lignocelulózu a bilirubin oxidáze z rostlinného patogenu Myrothecium verrucaria (MvBOx). X-ray crystallography enables determination of three-dimensional structures of biotechnologically and medically significant macromolecules which leads to a better understanding of their function. The high-resolution crystal structure of Chaetomium thermophilum FAD-dependent oxidoreductase reveals a unique His-Ser active-site pair located in an unusually large wide-open active site pocket with an uncommon preference for polyaromatic substrates. The structure–function studies of Myrothecium verrucaria bilirubin oxidase combined with mutagenesis of Trp396, the key residue of a unique covalent bond, revealed that various substrates use the residue primarily for binding and to a smaller extent for electron transfer.
