Abstract

Bakalářská práce se zabývá syntézou komplexů steroidních ligandů s přechodnými kovy, které by mohli najít uplatnění jako teranostické nosiče radionuklidů. Teoretická část obsahuje krátký přehled způsobů přípravy derivátů steroidních hormonů a jejich komplexů s přechodnými kovy včetně radioizotopů a cytotoxických vlastností těchto komplexů. V experimentální části je popsána syntéza steroidních komplexů se směsí izotopů 152Eu a 154Eu s ligandy odvozenými od konjugátů cholesterolu a stigmasterolu s tryptofanem a fenylalaninem. V práci byly studovány podmínky přípravy značených steroidních komplexů elektronickou autoradiografií. Bylo připraveno 8 steroidních komplexů s 152,154Eu. Posléze bylo provedeno značení azamakrocyklických ligandů 68Ga, eluovaným z komerčního radionuklidového generátoru IGG-100. Byly studovány podmínky značení čtyř vybraných ligandů: 1,4,7,10-tetraazacyklododekan-1,4,7,10-tetraoctové kyseliny, 1,4,7-triazacyklononan-1,4,7-trioctové kyseliny, 1,4,8,11-tetraazacyklotetradekan-1,4,8,11-tetraoctové kyseliny a 1,4,7-triazacyklononan-1,4,7-trimethylidenfosforné kyseliny v závislosti na pH a reakčním čase. Celkově byly připraveny čtyři komplexy [68Ga]DOTA, [68Ga]NOTA, [68Ga]TETA a [68Ga]TRAP.

This thesis deals with synthesis of complexes of transition metals with steroidal ligands, that could be potentially related to theranostic agents. Theoretical part of the project contains a short overview of the different synthetic approaches to derivatives of steroid hormones and their complexes with transition metals and their radioactive isotopes. Attention is also paid to cytotoxic activities of prepared compounds. Experimental part describes the synthesis of steroidal complexes of isotopic mixture 152,154Eu with ligands derived from the conjugates of cholesterol and stigmasterol with phenylalanine and tryptophan. Optimal reaction conditions for complexation were investigated by means of electronic autoradiography. Eight steroidal complexes with 152,154Eu were synthesized in this work. Later [68Ga]-labellling studies on azamacrocyclic ligands were carried out using 68Ga eluted from commercial radionuclide generator IGG-100. Favourable conditions for labelling azamacrocyclic ligands with 68Ga were studied on four different ligands: 1,4,7,10-tetraazacyclododecan-1,4,7,10-tetraacetic acid, 1,4,7-triazacyclononan-1,4,7-triacetic acid, 1,4,8,11-tetraazacyclotetradecan-1,4,8,11-tetraacetic acid and 1,4,7-triazacyclononan-1,4,7-trimethylidenphosphinic acid. Four [68Ga]-labelled compounds were prepared: [68Ga]DOTA, [68Ga]NOTA, [68Ga]TETA a [68Ga]TRAP.
