Abstract
Xylanázy spolu s ostatními fibrolytickými enzymy hrají klíčovou roli při degradaci celulózy v trávicím traktu býložravců i člověka a v dalších procesech anaerobní degradace biomasy v biotechnologiích a průmyslu. Tato diplomová práce se zaměřuje na charakterizaci xylanolytických enzymů bakterií rodu Butyrivibrio a Pseudobutyrivibrio. Purifikované aktivní frakce xylanáz byly elektroforeticky separovány na přístroji Protean II (BioRad) pomocí SDS-PAGE a vizualizovány metodou zymografie s přídavkem CM xylanu v polyakrylamidovém gelu. Bandy s výraznou aktivitou byly použity pro hmotnostní analýzu peptidových fragmentů (MALDI-TOF/TOF MS). Další charakterizace byla provedena metodou 2D-elektroforézy na polyakrylamidovém gelu s izoelektrofokusací prováděnou pomocí přístroje Protean IEF. Významná aktivita extracelulárních xylanolytických enzymů byla detekována u bakterií Butyrivibrio proteoclasticus, Pseudobutyrivibrio xylanivorans a Butyrivibrio fibrisolvens.
Xylanases and other fibrolytic enzymes play a crucial role in cellulose degradation important in ruminants or human gut and other processes of anaerobic degradation of biomass in biotechnology or industry. The aim of this diploma thesis is to identify and characterize xylanolytic enzymes of their significant producers of bacterial genus Butyrivibrio and Pseudobutyrivibrio. Purified active fractions of xylanases were electrophoretically separated on Protean II (BioRad) by SDS-PAGE and visualized by zymography with addition of CM xylan in polyacrylamid gel. Bands with significant activity were used for mass analysis of peptide fragment (MALDI-TOF/TOF MS). Further characterisation was carried out through 2D electrophoresis on polyacrylamid gel with isoelectrofocusation on Protean IEF (BioRad). Significant enzyme activity was detected in extracellular enzymes of Butyrivibrio proteoclasticus, Pseudobutyrivibrio xylanivorans and Butyrivibrio fibrisolvens.
