Abstract
Fotodynamická terapie indukovaná rentgenovským zářením je metoda léčby rakoviny kombinující radioterapii a fotodynamickou terapii, kde jsou nádorové buňky usmrceny singletovým kyslíkem. Ten může být produkován v nádorových tkáních vhodnými nanomateriály po ozáření rentgenovským zářením. Nanomateriály použitelné pro fotodynamickou terapii indukovanou rentgenovským zářením musí splňovat řadu požadavků pro využití v lékařství, přičemž jeden z požadavků je produkce singletový kyslíku a absence temné toxicity nanomateriálu. V této práci byla sledována temná toxicita nanomateriálu GGAG@SiO2-RB v různých stádiech funkcionalizace a toxicita po ozáření plně funkcionalizovaného nanomateriálu na kvasinkách Saccharomyces cerevisiae. Dále byla stanovena růstová křivka S. cerevisiae a zavedena spektrometrická metoda stanovení koncentrace buněk měřením na spektrofotometru jako náhrada počítání buněk pod mikroskopem s použitím Bürkerovy komůrky.
Photodynamic therapy induced by X-rays is an innovative method of cancer treatment combining radiotherapy and photodynamic therapy, in which cancer cells are killed by singlet oxygen produced by suitable nanomaterials in tumour tissues after exposure to X-rays. Nanomaterials usable for X-ray induced photodynamic therapy must meet a number of requirements for medical applications, one of which is the production of singlet oxygen and the absence of dark toxicity of the nanomaterial. In this work, the dark toxicity of the nanomaterial GGAG@SiO2-RB at different stages of functionalization and the toxicity after irradiation of the fully functionalized nanomaterial on yeast Saccharomyces cerevisiae were investigated. In addition, the growth curve of S. cerevisiae was determined and a spectrometric method was introduced to determine cell concentration by measuring on a spectrophotometer as a replacement of cell counting under a microscope using a Bürker chamber.
