Abstract
Cílem této studie bylo zkoumat toxicitu nanoˇcástic ZnO:Ga v°uˇci bakterii Escherichia coli DBM 3125 a její závislost na velikosti nanoˇcástic. V této studii byly použity nanoˇcástice s velikostí krystalit°u 7, 15, 33, 74 a 290 nm. (Velikost jejich krystalit°u byla mˇeˇrena pomocí rentgenové difrakce.) Bakterie byly vystaveny nanoˇcásticím po dobu 0,5; 1; 2; 3 a 4 h. Toxicita nanoˇcástic byla vyhodnocena na základˇe poklesu frakce pˇreživších bunˇek v ˇcase v porovnání s poklesem frakce pˇreživších bunˇek nevystavených nanoˇcásticím. Frakce pˇreživších bunˇek byla vypoˇcítána jako pomˇer poˇctu bunˇek v ˇcase t ku poˇctu bunˇek v ˇcase 0,5 h. Frakce pˇreživších bunˇek vystavených nanoˇcásticím klesala s ˇcasem. Na druhou stranu frakce pˇreživších bunˇek nevystavených nanoˇcásticím se v ˇcase témˇeˇr nemˇenila. To naznaˇcuje, že nanoˇcástice ZnO:Ga jsou toxické v°uˇci Escherichia coli DBM 3125. Toxicita nanoˇcástic ZnO:Ga mírnˇe rostla s rostoucí velikostí krystalit°u nanoˇcástic.
The aim of this paper was to research toxicity of nanoparticles ZnO:Ga against Escherichia coli DBM 3125 and its dependency on size of those nanoparticles. Nanoparticles with crystallites sizes 7, 15, 33, 74 and 290 nm were used. Size of those crystallites was measured by X-ray diraction. Bacteria were exposed to nanoparticles for durations of 0.5; 1; 2; 3 and 4h. Fraction of surviving cells was calculated as ratio of cell count in time t since exposure to nanoparticles to cell count at 0,5h. Toxicity of the nanoparticles was rated on basis of surviving cells in time t compared to the fraction of surviving cells not exposed to the nanoparticles. Fraction of surviving cells exposed to nanoparticles decreased with time. Fraction of cells not exposed to nanoparticles remained largely unchanged with time. This implies that nanoparticles of ZnO:Ga are toxic to Escherichia coli DBM 3125. Toxicity of nanoparticles ZnO:Ga slightly increased with the increase of crystallite nanoparticle size.
