Tato práca sa zameriava na problematiku digitálnej holografie a jej experimentálne použitie v digitálnej holografickej mikroskopii a tomografii. Práca je delená na dve časti. Prvá časť prace je rešeršného charakteru. Na začiatku podáva prehľad o holografii ako takej, predpokladoch pre záznam hologramu, geometrickom usporiadaní objektov a zahŕňa tiež úvod do skalárnej teórie difrakcie. Ta je východiskom pre numerickú rekonštrukciu digitálnych hologramov, ktorým sa venuje ďalšia kapitola. V nej sú následne diskutovane metódy odstránenia nultého difrakčného rádu a zdvojeného obrazu s podrobnejším zameraním sa na techniku posunu fázy. Tiež sú priblížene holografické konfigurácie digitálnej holografie, používane numerické techniky a niektoré jej aplikácie, ako digitálna holografická mikroskopia a tomografia. Tieto dve aplikácie sú predmetom experimentálnej druhej časti prace, v ktorej sú demonštrovane možnosti kvantitatívnej rekonštrukcie poľa, obsahujúcom informácie o zmenách fázy, resp. indexu lomu vo vzorkách. The thesis focuses on the topic of digital holography and its experimental application in digital holographic microscopy and tomography. The thesis consists of two parts. The first part contains background research. At the beginning there is an overview of the holography as a whole, premises for holographic recording and geometric layout of objects. An introduction to scalar theory of diffraction is included as well. The scalar theory represents the basis for numerical reconstruction of the digital holograms, which are discussed in the next chapter. Consequently, the methods for removing the zero order and twin image with an emphasis on phase-shifting methods are also examined. The further points discussed include digital holography configurations, numerical techniques and some of its applications, such as digital holographic microscopy and tomography. These two applications are the subject of the second experimental part of the thesis. There the possibilities of quantitative field reconstruction containing information about changes of phase and refractive index in sample volume are demonstrated.
