Analýza mechanických vlastností buněk umožňuje nový pohled na změny jejich chování vlivem prostředí, ve kterém se nacházejí a na změny jejich struktury, které mohou být způsobené patologickými změnami. Zejména struktura buňky v podobě cytoskeletu, cytoplazmatické membrány a extracelulárního matrixu má velký vliv na mechanické vlastnosti. Následně patologie buněk může tuto strukturu narušit a způsobit změny v jejím biomechanickém chování. Tato práce je zaměřena na měření mechanických vlastností buněk pomocí mikroskopie atomárních sil (AFM) neboli metody zvané nanoindentace. Cílem práce je zpracování teoretické rešerše z předešlých studií a zaměření na zhotovení nástroje, který bude schopný vyhodnocovat mechanické vlastnosti buněk, a to konkrétně Youngův modul pružnosti, pomocí matematických modelů jako je Hertzův model kontaktních tlaků a Oliver Pharr model. Dále je práce zaměřena na automatické separování výsledků z jednotlivých částí buňky jako je jádro, cytoplazma a její okolí. Výsledkem práce je automatizace, urychlení práce při zpracovávání naměřených dat a porovnání výsledků. Konkrétní výsledky byly sepsány v závěru práce. Analysis of the mechanical properties of cells provides new insight into changes in their behaviour due to their environment and changes in their structure that may be caused by pathological changes. In particular, the structure of the cell in the form of the cytoskeleton, cytoplasmic membrane and extracellular matrix has a major influence on mechanical properties. Consequently, cell pathology can disrupt this structure and cause changes in the biomechanical behaviour of the cell. The aim of this thesis is to elaborate on the theoretical research from previous studies and focus on the fabrication of a tool that will be able to evaluate the mechanical properties of cells, specifically the Young's modulus of elasticity using mathematical models such as the Hertz contact pressure model and the Oliver-Pharr model. Furthermore, the work is aimed at automatically separating the results from different parts of the cell such as the nucleus, cytoplasm and its surroundings. The work results in automation, speeding up the processing of measured data and comparison of results. The specific results were written in the conclusion of the paper.
