Tato bakalářská práce se zabývá problematikou intenzivně zkoumaných antifosfolipidových protilátek a jejich aviditou. Přítomnost antifosfolipidových protilátek způsobuje antifosfolipidový syndrom charakterizovaný trombózami a komplikacemi v těhotenství. Teoretická část obsahuje aktuální informace o typech antifosfolipidových protilátek, kritériích pro diagnostiku antifosfolipidového syndromu a je vysvětlen pojem avidita a afinita. V praktické části bylo provedeno a popsáno stanovení avidity antifosfolipidových protilátek, konkrétně antifosfatidylethanolaminových protilátek (aPE). Ke stanovení byla použita imunologická metoda ELISA, za použití různých chaotropních činidel, konkrétně urey a NaCl. Byla testována různá kontrolní směsná séra, intravenózní imunoglobuliny a vzorky pacientů. Porovnáním výsledků byla z použitých chaotropních činidel vyhodnocena urea jako vhodnější. Ve všech případech stanovení se nám podařilo zachytit klesající trend indexu avidity se stoupající koncentrací chaotropního činidla. Výsledky také naznačily, že u pacientů trpících trombózami by se mohly vyskytovat aPE s vyšší aviditou v porovnání se zdravými osobami. This bachelor thesis deals with the issue of intensively researched antiphospholipid antibodies and their avidity. The presence of antiphospholipid antibodies causes the antiphospholipid syndrome characterized by thromboses and complications in pregnancy. The theoretical part contains current information about the types of antiphospholipid antibodies, criteria for the diagnosis of antiphospholipid syndrome and the concept of avidity and affinity is explained. In the practical part, the determination of the avidity of antiphospholipid antibodies, specifically antiphosphatidylethanolamine antibodies (aPE), was performed and described. The immunological ELISA method was used for the determination, using different chaotropic reagents, namely urea and NaCl. Various control mixed sera, intravenous immunoglobulins and patient samples were tested. By comparing the results, we evaluated urea as the more appropriate of the chaotropic reagents used. In all cases of determination, we were able to detect a decreasing trend of avidity index with increasing concentration of chaotropic reagent. The results also indicated that aPE with higher avidity is found in patients suffering from thromboses.
