Probiotické bakterie mají spousty pozitivních dopadů na lidský organismus. Bakterie samotné nebo jejich metabolity jsou schopny stimulovat náš imunitní systém a genovou expresi cytokinů. Většina výzkumů probiotik v dnešní době se zaměřuje na jejich využití v léčbě astmatu, alergie a jejich působení proti zánětlivým onemocněním. Tato práce poskytuje přehled o nejznámějších probiotických připravcích a jejich vlivech na imunitní systém. Pro lepší pochopení působení bakterie E. coli a jejich metabolitů na lidský imunitní systém byly testovány její imunomodulační vlastnosti na izolovaných buňkách mononukleární frakce z buffy coatu. Konkrétně byla sledována schopnost různých koncentrací metabolitů, produkovaných v anaerobní a aerobní kultivaci E. coli stimulovat hladiny vybraných zánětlivých a prozánětlivých cytokinů. Všechny hladiny cytokinů byly měřeny pomocí polymerázové řetězové reakce. Výsledkem této práce jsou grafy, ukazující hladiny cytokinů po přidání stimulátorů v porovnání s nestimulovanými buňkami. Je patrné, že metabolity z anaerobní kultivace E. coli AN 200 a AN 40 zvyšují genovou expresi Il10 a metabolity z aerobní kultivace E. coli AE 200 a AE 40 zvyšují genovou expresi Ifng a Il2. Tyto zmíněné cytokiny IL-10, IL- 2 a IFNg podporují Th1 imunitní odpověď, která hraje důležitou roli při autoimunitních odpovědích. Z tohoto můžeme usuzovat, že zmíněné metabolity by mohly být v budoucnu použity v probiotických produktech na léčbu zánětů (hlavně trávícího traktu) a alergií. Probiotic bacteria have positive impact on the human body. Bacteria themselves or their metabolites are able to stimulate our immune system and cytokine gene expression. Most research on probiotics today focuses on their use in the treatment of asthma, allergies and their action against inflammatory diseases. This work provides an overview of the most well-known probiotic vaccines and their effects on the immune system. To better understand the effects of E. coli and their metabolites on the human immune system, its immunomodulatory properties were tested on isolated cells of the mononuclear fraction from the buffy coat. Specifically, the ability of various concentrations of metabolites produced in anaerobic and aerobic culture of E. coli to stimulate the levels of selected anti-inflammatory and proinflammatory cytokines was studied. All cytokine levels were measured by polymerase chain reaction. The result of this work are graphs showing cytokine levels after the addition of stimulators in comparison with unstimulated cells. It can be seen that metabolites from E. coli anaerobic culture AN 200 and AN 40 upregulate Il10 gene expression and metabolites from aerobic E. coli culture AE 200 and AE 40 upregulate Ifng and Il2 gene expression. Cytokines IL-10, IL-2 and IFNg support the Th1 immune response, which plays an important role in autoimmune responses. From this we can conclude that the metabolites tested could be used in the future in probiotic supplements for the treatment of inflammation (mainly of the digestive tract) and allergies.
