Předmětem této bakalářské práce bylo stanovit referenční rozmezí agregace destiček pro pacienty s případným tzv. syndromem lepivých destiček. Pro získání výsledných hodnot referenčního rozmezí byla proměřena skupina 40 zdravých dobrovolníků za použití Bornovy optické transmisní agregometrie (Light Transmission Aggregometry – LTA). Byla stanovena indukovaná agregace destiček po přidání induktorů agregace epinefrinu (EPI) a adenosinu difosfátu (ADP) ve snížených koncentracích. Hodnoceným parametrem byla maximální agregace v %. Rozdělení naměřených hodnot nevyhovovalo podmínkám normality dle Gaussovy křivky, a tak bylo referenční rozmezí vyjádřeno přímou neparametrickou metodou. Referenční rozmezí je pro dílčí koncentrace induktorů následující: EPI (konc. 1,0 μmol/l) = 2–77 %; EPI (konc. 0,1 μmol/l) = 1–19 %; EPI (konc. 0,05 μmol/l) = 1–5 %; ADP (konc. 2,34 μmol/l) = 19–76 %; ADP (konc. 1,17 μmol/l) = 4–65 %; ADP (konc. 0,58 μmol/l) = 1–11 %. Podle stanoveného referenčního rozmezí je možné diagnosticky hodnotit výskyt syndromu lepivých destiček u pacientů s doposud neobjasněným zvýšeným sklonem k trombózám v mladším věku, s hemodialyzační léčbou, u pacientů po transplantaci ledviny nebo u pacientek, které prodělaly několikanásobné aborty. The aim of this bachelor thesis was to establish a reference range of platelet aggregation for patients with possible Sticky Platelet Syndrome. A group of 40 healthy volunteers was studied using Born's Optical Transmission Aggregometry (LTA) to obtain the resulting reference range values. Induced platelet aggregation was determined after addition of the aggregation inducers epinephrine (EPI) and adenosine diphosphate (ADP) at reduced concentrations. The evaluated parameter was the maximum aggregation in %. The distribution of the measured values did not satisfy the normality conditions according to the Gaussian curve, so the reference range was expressed by the direct non-parametric method. The reference ranges for the sub-concentrations of the inducers are as follows: EPI (conc. 1.0 µmol/l) = 2–77 %; EPI (conc. 0.1 µmol/l) = 1–19 %; EPI (conc. 0.05 µmol/l) = 1–5 %; ADP (conc. 2.34 µmol/l) = 19–76 %; ADP (conc. 1.17 µmol/l) = 4–65 %; ADP (conc. 0.58 µmol/l) = 1–11 %. According to the established reference range, the incidence of sticky platelet syndrome can be diagnostically assessed in patients with a previously unexplained increased propensity to thrombosis at a younger age, with haemodialysis treatment, in patients after renal transplantation or in patients who have had multiple abortions.
