dc.description.abstract | Diplomová práce pojednává o vlivu různých degradačních médií, simulujících prostředí lidského těla, na mechanické vlastnosti kolagenních nosičů. K tomuto účelů byla vybrána tři degradační média: lidská plazma, simulovaná tělní tekutina (SBF) a fosfátový pufr (PBS). Cílem provedeného experimentu bylo zjistit, zda jejich aplikací bude dosaženo stejného degradačního efektu, či nikoli. Experiment byl proveden na nosičích z kolagenu typu I chemicky zesíťovaných roztokem 95% ethanolu, N- (3-dimethylaminopropyl) -N-ethylkarbodiimid hydrochloridu (EDC) / N-hydroxysukcinimidu (NHS). V první části experimentu byly nosiče naloženy do příslušného média a ponechány jeho působení po zvolené časové intervaly: 0 dnů (vzorky byly hydratovány 10 minut před mechanickým testem), 7 dní a 14 dní. Mechanické vlastnosti byly vyhodnoceny axiální tlakovou zkouškou. Druhá část experimentu byla zaměřena na posouzení hmotnostního úbytku kolagenních nosičů. Nosiče byly působení roztoků vystaveny opět po 7 a 14 dní. I přesto, že roztoky simulují stejné prostředí a s krevní plasmou mají podobnou iontovou koncentraci, ukázalo se, že každý z použitých roztoků má odlišný vliv na vývoj mechanických vlastností i na hmotnostní úbytek. | cze |
dc.description.abstract | Thesis discusses the topic of collagen scaffolds degradation, particularly the influence of different degradation media, simulating the human body environment, on mechanical properties of collagen scaffolds. There were three degradation media used: human plasma, Simulated Body Fluid (SBF) and Phosphate Buffered Saline (PBS). The aim of the following experiment was to determine whether the degradation effect of the media will be the same for all used media, or not. Scaffolds were composed of collagen type I isolated form calf skin and were chemically crosslinked with the solution of 95% ethanol, N-(3-dimethylaminopropyl)-N-ethylcarbodiimide hydrochloride (EDC) / N-hydroxysuccinimide (NHS). In the first part of the experiment, scaffolds were subjected to the degradation effect of given media for selected time periods: 0 days (samples were hydrated 10 minutes before mechanical test), 7 days and 14 days. Mechanical properties were examined by axial compression test. The second part of the experiment focused on the determination of the scaffold weight loss. Samples were exposed to the media for 7 and 14 days. Although the solutions simulate the same environment and have a similar ionic concentration as blood plasma, it has been shown that each of the solutions used has a different effect on the development of mechanical properties and on weight loss. | eng |
