Předkládaná diplomová práce se zabývá sledováním genotoxickýchúčinků mutagenů N- nitroso-N-metylurey (MNU), aflatoxinu B1 (AFB1), 2-amino-3-metylimidazo[4,5-f]chino- linu (IQ), bleomycinu, a antimutagenníchúčinků lykopenu a kurkuminu. In vivometodou mikronukleus testu na myších erytrocytech byl prokázán statisticky významnýúčinek kurkuminu a lykopenu na snížení mutagenity přímého mutagenu MNU a nepřímých mutagenů AFB1 a IQ.Účinek obou látek byl výraznější proti nepřímým mu-tagenům. Součástí práce bylo zavedeníin vitrometody mikronukleus testu na lidských pe-riferních lymfocytech, kterou byly sledovány antimutagenníúčinky kurkuminu vůči MNU. Počet mikrojader indukovaných MNU byl dávkově závislý. Bylo pozorováno pouze statis- ticky nevýznamné snížení mutagenity MNU kurkuminem o vyšší koncentraci (20 ?g/ml). Dále byly sledovány mutagenníúčinky MNU na indukci chromozomálních aberacív lidských periferních lymfocytech. Výskyt aberací byl dávkově závislý, ve všech koncen- tracích převažovaly chromatidové aberace, nejčastější byly chromatidové zlomy. Studie typů chromozomálních aberací indukovaných bleomycinem prokázala závislost na fázi buněčného cyklu a na koncentraci mutagenu. Bleomycin přidaný na 5 hodin na začátku kultivace indukoval převážně chromozomové aberace. Bleomycin přidaný na 5 hodin po 24 hodinách kultivace ve vyšší koncentraci (30?g/ml) způsobil více chromozomovýchaberací, zatímco pro nižší koncentraci (3?g/ml) bylo více chromatidových aberací. Popůsobení bleomycinu v posledních 5 hodinách kultivace byly téměř všechny aberace chro- matidové a počet aberantních buněk a aberací byl nejvyšší. The present thesis is focused on the study of genotoxic effects of mutagens N-nitroso-N- metylurea (MNU), aflatoxin B1 (AFB1), 2-amino-3-metylimidazo[4,5-f]chinolin (IQ) and bleomycin and the study of anti-mutagenic effects of lycopene and curcumin. In vivomethod of micronucleus test on mice erythrocytes showed a statistically sig- nificant effect of curcumin and lycopene on the reduction of the mutagenicity of direct mutagen MNU and indirect mutagens AFB1 and IQ. The effect of both substances was more pronounced against indirect mutagens. The part of this work was aimed to the im- plementation ofin vitromethod of micronucleus assay on human peripheral lymphocytesfor the detection of antimutagenic effects of curcumin against mutagenicity of MNU. The number of micronuclei induced by MNU was dose-dependent. It has been observed only a statistically insignificant reduction of mutagenicity MNU by curcumin used at higher concentration (20?g/ml). Mutagenic effect of MNU on the induction of chromosomal aberrations in human peripheral lymphocytes was also investigated. The incidence of aberrations was dose- dependent, chromatid aberrations prevailed at all concentrations used, the most frequent aberrations were chromatid breaks. The study of chromosomal aberrations induced by bleomycin revealed that the number and types of chromosomal aberrations are related to the cell cycle phase and to the mutagen concentration. Bleomycin added for 5 hours at the beginning of cultivation induced predominantly chromosome aberrations. Bleomy- cin added for 5 hours after 24 hours of cultivation at higher concentration (30 ?g/ml)caused more chromosome aberrations, whereas the lower concentration (3?g/ml) indu-ced prevalence of chromatide aberrations. After bleomycin exposure for the last 5 hours of cultivation almost all aberrations were of chromatid type and the number of aberrant cells and aberrations was the highest.
