Pro určování vývoje a stáří živočichů je stále standardně využíváno histologických a mikroskopických metod. Tato práce ukazuje na možnosti využití řádově méně náročné, nedestruktivní mikroradiografie a mikrotomografie. Cílem práce je navrhnutí a provedení radiografického měření pro aplikace ve vývojové morfologii, která se zabývá vývojem stavby živočichů. Byly nalezeny a otestovány tři aplikace. Nejprve byly pomocí mikroradiografie sle-dovány růstové chrupavky pro potvrzení č?i vyvrácení obecně uznávaného předpokladu neukončeného růstu plazů. Dále byly mikrotomografickou metodou znázorněny kostní přírůstky, díky kterým jsme schopni odhadnout stáří zvířete, a které jsou standardně zobrazovány histologickou metodou. V posledním experimentu je ukázáno na výhodu radiografických metod, kde můžeme využít naměřená data pro znázornění například trabekulární struktury kosti. Výsledkem diplomové práce je aplikace mikroradiografie a mikrotomografie ve vývojové morfologii. Na základě výsledků měření dochází ke zpochybnění teorie neukončeného vývoje plazů, zejména u menších druhů ještěrů. Dále je ukázáno, že kostní přírůstky jsou pomocí mikroradiografie znázornitelné. Z provedených experimentů vyplývá, že v aplikacích v oblasti vývojové morfologie má mikroradiografie a mikrotomografie svůj potenciál. Histologic and microscopic methods are still widely used for determin-ing the age and evolution of the animals. This study demonstrates the possibil- ity of using less time consuming, non-destructive microradiography and microtomography methods. The aim of diploma thesis is to propose and perform radiographic meas-urements for application in developmental morphology, which is engaged in the development of structure of the animals. Three methods were found and tested as well. The first, the growth plates of reptiles were monitored by using microradiography to confirm or disprove the generally accepted assumption of indetermined growth in reptiles. In the next measurement, the skeletal cyclic growth marks were shown, thanks to which we are able to esti- mate the age of the animal, which are normally shown by histological method. In the latest experiment the advantage of radiographic methods is presented where we can use previously measured data to illustrate for example the tra- becular structure of the bone. The result of the thesis is application of microradiography and microto-mography in the field of developmental morphology. Based on the results, the theory of indetermined growth in reptiles, especially in smaller species of lizards was disproof. It is shown that cyclic growth marks can be represented by using microtomography. The experiments prove that microradiography andmicrotomography could replace histology in applications in the field of developmental morphology.
