Stanovení funkce trombocytů pomocí impedanční agregometrie

Abstract

Predmetem teto prace je stanovit referencn rozmez u agregometrickeho vysetren funkce trombocytu u zdravych darcu krve. Referencn rozmez u agregometrickeho vysetren funkce trombocytu jsme stanovili pomoc kontroln skupiny (zdravych darcu krve), u kterych jsme vyhodnocovali jednotlive induktory agregace trombocytu (ADP test, ASPI test a TRAP test). Namerena data jednotlivych induktoru jsme statisticky zpracovali a pomoc vypoctu prumeru, smerodatne odchylky a 1,96nasobku smerodatne odchylky jsme urcili referencn rozmez u kontroln skupiny, ktere jsme pote porovnali s vyrobcem analyzatoru Multiplate, Roche, a dalsmi dostupnymi zdroji. V dals casti teto prace se zabyvame s pomoc impedancn agregometrie stanovenm ucinnosti lecby u pacientu na antiagregacn lecbe uzvajcch clopidogrel nebo kyselinu acetylsalicylovou (ASA). Ucinnost lecby clopidogrelem aASA je vyjadrena jako procentualn inhibice agregace, kterou jsme spoctali pomoc vzorcu pro vypocet mry inhibice agregace.

The subject of this thesis is to specify the reference range for platelet function testing by aggregometry. The data used for this task come from healthy blood donors. The reference range for platelet function testing by aggregometry was specied using control group (healthy blood donors). We have made platelet aggregation measurements of various inductors (ADP test, ASPI test and TRAP test) for the control group. We have statistically processed the measured data.We have calculated the mean value, standard deviation and 1.96 times standard deviation and used it for dening the reference range. We have made comparisons between our reference ranges and those proposed by the producer of Multiplate Analyzer, Roche, and other available sources. Next part of this thesis is concerned with determination of ecacy of the treatment of patients who use antiplatelet drugs á clopidogrel or acetylsalicylic acid (ASA). The method used for measuring the ecacy was impedance aggregometry. The ecacy of the treatment with clopidogrel and ASA is expressed as percentage inhibition of platelet aggregation. We have calculated the ecacy with the aid of formula for calculation of inhibition of platelet aggregation.
