Jednou z metod asistované reprodukce je takzvaná in vitro fertilizace. Vajíčka jsou oplodněna mimo tělo ženy a vzniklá embrya jsou poté uložena v kultivačním prostředí uvnitř inkubátoru. Růst a vývoj embryí je následně přibližně 5 dní pozorován a nejkvalitnější embrya jsou následně přenesena zpět do dutiny děložní. Práce má za cíl prozkoumat možnosti využití digitální holografické mikroskopie ke sledování růstu embryí uvnitř inkubátoru. Práce samotná je rozdělena na část teoretickou a praktickou. V teoretické jsou nejprve představeny principy digitální holografické mikroskopie a 2 druhy sestav mikroskopů. Následně jsou představeny současné aplikace a komerční holografické mikroskopy. Poté následuje popis matematického aparátu použitého k numerické rekonstrukci obrazu. V praktické části je představen postup návrhu a realizace jednotlivých experimentálních sestav digitálního holografické mikroskopu. Poté je představen program pro rekonstrukci obrazu. Následuje popis metodiky záznamu hologramu a vyhodnocení rekonstruovaného obrazu z experimentálních sestav. Nakonec jsou představeny výsledky v podobě vybraných rekonstruovaných hologramů, popisu parametrů mikroskopu a návrhu sestavy pro práci v inkubátoru. One of the methods of assisted reproduction is the so-called in vitro fertilization. The eggs are fertilised outside the woman's body and the resulting embryos are then stored in a culture environment inside an incubator. The growth and development of the embryos is then observed for approximately 5 days and the best quality embryos are then transferred back into the uterine cavity. This thesis aims to investigate the possibilities of using digital holographic microscopy to monitor embryo growth inside the incubator. The thesis itself is divided into theoretical and practical parts. In the theoretical part, firstly the principles of digital holographic microscopy and 2 types of microscope assemblies are introduced. Subsequently, current applications and commercial holographic microscopes are introduced. This is followed by a description of the mathematical tools used for numerical image reconstruction. In the practical part, the design and implementation procedure of the different experimental setups of the digital holographic microscope is presented. Then the image reconstruction program is presented. This is followed by a description of the hologram recording methodology and the evaluation of the reconstructed image from the experimental setups. Finally, the results are presented in the form of selected reconstructed holograms, a description of the microscope parameters and the design of the assembly for work in the incubator.
