V této práci jsou provedeny simulace laserového plazmatu pomocí rozšířeného magnetohydrodynamického modelu (ExMHD). K numerickým simulacím interakcí mezi laserového záření s terčem ve vnějším magnetickém poli je použit kód FLASH. Je posouzen vliv a fyzikální význam jednotlivých procesů zahrnutých v modelu ExMHD. Modely transportu magnetizovaného plazmatu jsou důkladně prostudovány simulacemi jednotlivých jevů. Je představeno numerické schéma, založené na metodě konečných prvků, pro Nernstův efekt a je zkoumána vazba mezi termoelektrickými členy. Do ExMHD modelu je přidán proud horkých elektronů, který vede k vybuzení magnetického pole na čele plazmatické koróny. Toto pole je porovnáno se spontánním magnetickým polem vyvolaným Biermannovým efektem.; In this thesis, various simulations of laser-produced plasma are performed using the extended magnetohydrodynamic (ExMHD) model. The FLASH code is utilized to perform numerical simulations of laser-target interactions in an external magnetic field. The impact and physical meaning of individual effects included in the ExMHD is assessed. The magnetized plasma transport models are studied thoroughly in simulations focusing on single phenomena. A finite element numerical scheme for the Nernst effect is presented and coupling between the thermoelectric contributions is examined. Hot electron current is added into the ExMHD model, which leads to the generation of an angular magnetic field at the plasma corona front. This field is compared to the cross-gradient magnetic field induced by the Biermann effect.

V posledních letech došlo v oblasti ultracitlivé mikroskopie k významnému pokroku v citlivosti a kvantitativní povaze měření. Zejména pro pochopení vnitřních struktur opticky tenkých objektů, jako jsou buňky a tkáně, se fázová citlivost stává klíčovou. Tato práce zkoumá nové funkce a implementace v interferometrické rozptylové mikroskopii (iSCAT), která je proslulá svými neinvazivními zobrazovacími schopnostmi bez použití značek. iSCAT je díky své citlivosti na změny fáze ideální metodou pro ultracitlivou detekci fáze, což je klíčový aspekt při pozorování dynamiky jednotlivých molekul. Tato studie demonstruje, jak iSCAT dokáže kvantitativně detekovat fázi rozptýleného světla nezávisle na jeho amplitudě, s cílem zvýšit přesnost kvantifikace makromolekulárních interakcí. Výzkum dále zkoumá, jak fáze ovlivňuje iSCAT kontrast, a ukazuje, že i subnanometrové nerovnosti krycích sklíček mohou významně ovlivnit výsledný interferogram. Tato zjištění nejen zdůrazňují potenciál iSCATu v molekulární biologii, ale také poukazují na nutnost přesnosti v mikroskopickém prostředí, což otevírá cestu k přesnějším a spolehlivějším mikroskopickým analýzám.; In recent years, the field of ultrasensitive microscopy has seen significant advancements in its sensitivity and quantitative nature of the readout. Particular for understanding the internal structures of optically thin objects like cells and tissues, quantitative phase sensitivity becomes crucial. This thesis explores new features and implementations in interferometric scatterring microscopy (iSCAT), renowned for its non-invasive label-free imaging capabilities. iSCAT’s sensitivity to phase changes makes it an ideal method for ultrasensitive phase detection, a pivotal aspect in observing the dynamics of single molecules. This study demonstrates how iSCAT can quantitatively detect the phase of scattered light independent of its amplitude, aiming to advance the accuracy in quantification of macromolecular interactions. The research further investigates how the phase affects iSCAT contrast and reveals that even subnanometer unevenness in coverslips can significantly influence the resulting interferogram. These findings not only underscore the potential of iSCAT in molecular biology but also highlight the necessity for precision in microscopic environments, paving the way for more accurate and reliable microscopic analyses.

Ultra krátké pulzy, obvykle trvající desítky nebo stovky femtosekund jsou klíčovým nástrojem pro výzkum elektronové dynamiky. Nicméně některé pokročilé aplikace vyžadují ještě kratší pulzy. Few-cycle pulzy, sestávající se pouze z 2-3 period tomuto vyhovují, avšak mají i další výhody. Jejich hlavní výhoda spočívá v tom, že špičková intenzita je dosažena téměř okamžitě, v průběhu jednoho optického cyklu. Tím umožňují například efektivnější generaci vysokých harmonických, nebo využít takzvaný carrier-envelope offset, čímž je zvýšena míra kontroly na pulzem. Fundamentální překážka nám však brání v generování few-cycle pulzů přímo laserem. Nejkratší možná délka pulzu je dána jeho spektrální šířkou, která je zase převážně určena aktivním médiem laseru. Aby bylo možné toto omezení překonat, je nutné nejdřív rozšířit spektrum. Toho lze dosáhnout zpětně, poté co pulz opustí laser, a to pomocí nelineární interakce s médiem. To je základní idea, na které stojí technika post komprese. V této práci připravujeme experimentální sestavu pro post kompresi s cílem generovat few-cycle pulzy. Návrh experimentu počítá s využitím dvou různých nelineárních jevů pro rozšíření spektra, a sice ionizaci optickým polem a Kerrovým jevem indukovanou automodulaci. Cílem je sestavit zařízení, které bude schopné se vypořádat s extrémně vysokým špičkovým výkonem v řádu terawattů.; Ultra-short pulses, typically with duration in tens or hundreds of femtoseconds, are the key tool for the research of electron dynamics. However, in some cases, even shorter pulses are required. Few-cycle pulses, consisting of only 2-3 periods, satisfy this requirement, however, they have more to offer apart from being extremely short. The main advantage is linked to the fact that the peak intensity is reached almost immediately, over the course of a fraction of an optical cycle. This allows for example to generate HHG more efficiently, or exploit carrier-envelope offset, exerting additional control over the pulse properties. However, a fundamental obstacle prevents us from generating few-cycle pulses from the laser directly. The shortest possible duration of a pulse is given by its bandwidth, which in turn is given mostly by the active medium of the laser. One would first need to broaden the pulse spectrum to overcome this limitation. That can be achieved ex post, after the pulse leaves the laser, via non-linear interaction with a medium. This is the fundamental idea behind the post-compression technique. In this work, we prepare a setup for a post-compression experiment, with the aim to generate few-cycle pulses. The experiment is designed to exploit two different non-linear phenomena for spectral broadening purposes, optical field ionization, and Kerr effect-induced self-phase modulation. The goal is to build a device able to deal with extremely high peak power in the terawatt region.

Tato diplomová práce se zabývá optimalizací polovodičových jednofotonových detektorů, tyto typy detektorů mohou dosáhnout časového rozlišení lepšího než 20 ps a stability zpoždění výrazně lepší než 1 ps. Používají se v mnoha oblastech vědy a průmyslu nejen na Zemi, ale i ve vesmíru. Hlavním zájmem této diplomové práce je detektor vyvíjen pro ''European Laser Timing'', což je projekt Evropské vesmírné agentury, který má za cíl zlepšit přenos informace o času z atomových hodin, které jsou na palubě Mezinárodní vesmírné stanice, na Zemi. Cílem této diplomové práce je zlepšit teplotní stabilitu zpoždění detektoru a rychlost jeho zhášení, čehož jsem se snažil dosáhnout modernizací současné podoby řídicího obvodu detektoru. Tedy jsem vhodně upravil celé detektorové pouzdro a změřil jeho nové vlastnosti. Nový detektor vykazoval jak lepší stabilitu detekčního zpoždění, tak rychlost zhášení, proto jsem usoudil, že cíle diplomové práce byly splněny. Nakonec tato práce pokračovala nad rámec původního zadání a zkoumala také vylepšení kontinuálně běžících detektorů jednofotonových detektorů u kterých jsem srovnával různé konfigurace jejich řídícího obvodu.; This master thesis deals with optimization of semiconductor single photon detectors, these kinds of detectors can achieve time resolution better than 20 ps and propagation delay stability significantly better than 1 ps. They are used in many areas of science and industry, not only on Earth, but also in space. The detector that is of main interest to this master thesis is being developed for European Laser Timing, which is a European Space Agency project meant to facilitate transmission of time information from the atomic clock aboard International Space Station to Earth. The goals of this master thesis are to improve the detector's propagation delay temperature stability and the detector's quenching speed. I have attempted to accomplish this by modernizing the current design of the detector's control circuit. I have appropriately modified the integrated detector package and measured its novel properties. The new detector package displayed both better propagation delay stability and quenching speed, therefore I have concluded this master thesis as successful. In the end, this work proceeded beyond the scope of the thesis assignment and explored the improvement of continuously operated single photon detectors, for which I have compared different configurations of their control circuits.
